    Procedure to Use Microscope
    Before you use a microscope, there are several things you must consider, among other are as follows
  1. a. The microscope must be carried with two hands. The left hand up holds (is placed under) the microscope and right hand holds the microscope arm.
  2. b. When observing wet preparation, the microscope body must be in upright position or the preparate desk must be in a flat position
  3. c. Wet preparationmust always be covered with a covering glass.
  4. d. Keep the microscope lenses and mirror clean
  5. e. Before using the microscope, you should check it first. If there is a microscope part which is broken, not working well, or if its objective lense is missing, then immediately report it to the teacher.
  6. f. During practiculum, you must not remove any of the objective lenses.
  7. g. When the practiculum activity is finished, the objective lens in the microscope that has been used must be rotated again to the lowest magnification (in straight downward position)
As for the methot to use microscope to observe an object. It is as follows.
  1. a. Put the microscope on the practicum table. Put on an ocular lens with weak magnification, for example 10x. (Commonly ocular lens has been in weak magnification, so we need not replace it)
  2. b. Rotate the macrometer backward, so the microscope can be lifted.
  3. c. Shift the lens rotation so the objective lens with weak magnification is in line with the direction of the incoming light. In the beginning
  4. d. Of microscope use, select objective lens with weak magnification for example 10x. So image magnification of 10x10=100 is obtained
  5. e. Lift the condenser as hight as possible. Open the diaphragm as wide as possible to allow enogh light to enter the condenser, Observe the object by 100x magnification.
  6. f. Put Preparation on the hole of preparation desk. So that the preparation is passed by the light from condenser. Hold the preparation with a clamp.
  7. g. Rotate the macrometer forward unti the objective lens is exactly on the observed object
  8. h. Lift and lower the microscope body by moving the macrometer while adjusting the focus connection the ocular lens to objective lens until the preparation appears clearly.
  9. i. Observe object by magnification of 100x.
  10. j. Move the lens rotator until the objective lens with 45x magnification is the right position, as marked by a clicking sound.
  11. k. While observing the object, move the micrometer forward or back-ward until the preparation appears clearly.
  12. l. Observe the preparation and if necessary make a drawing of it.
  13. m. If you want to observe bacteria preparation, special treatment is needed, that is by using immersion oil which is dropped on the preparation. This is done by first moving away the objective lens from preparation. This is done by first moving away the objective lens from preparation. Rotate the objective lens with magnification of 100x and move it down until touching the immersion oil.
  14. n. If observation is finished, rotate the objective lens of high magnification lens.
  15. o. Lift the microscope body or lower the object desk by using the micrometer, after that release the preparation from object desk.
